BioCina announces opening of new GMP plasmid DNA manufacturing suite

BioCina, a leading contract development and manufacturing organization (CDMO) today announced that the company has expanded its CDMO service offering to include the manufacture of plasmid DNA (pDNA) in a new dedicated GMP suite. The suite is equipped with up to 300 L single use fermentation capacity and suitably scaled downstream processing equipment. To complement GMP manufacturing, a broad range of analytical methods have been developed to support in-process, release and stability testing of pDNA products. This is a very important and timely addition that will best enable BioCina to work with its industry partners to further develop RNA vaccines and therapeutics as well as cell and gene therapies.

BioCina's decision to expand its service offering into pDNA development and manufacturing was driven by the global increase in demand for GMP grade pDNA fueled by the exponential growth in vaccines and therapies on the RNA platform as well as strong growth in the cell and gene therapy markets.  These factors, combined with the CDMO industry's dearth of capacity and access to high-quality cGMP pDNA manufacturing expertise, made this a clear path forward underscored by BioCina's depth of experience in microbial manufacturing.

Tempus Announces Companion Diagnostic Collaboration with TScan Therapeutics

Tempus, a leader in artificial intelligence and precision medicine, announced a new collaboration to develop a companion diagnostic (CDx) test with TScan Therapeutics, a clinical-stage biopharmaceutical company focused on the development of T cell receptor (TCR)-engineered T cell therapies (TCR-T) for the treatment of cancer patients. The collaboration supports TScan’s screening protocol for its Phase 1 solid tumor clinical trial which is designed to enable customized mixtures of TCR-Ts to be administered to patients based on tumor antigen positivity and intact HLA expression. TCR-Ts genetically reprogram a patient’s immune system to recognize and fight their cancers. TScan plans to enroll patients with solid tumors including non-small cell lung cancer, melanoma, head and neck cancer, ovarian cancer, and cervical cancer. Up to 40% of these tumors lose half of their HLA genes, which is a frequent and overlooked cause of resistance to immunotherapies such as TCR-Ts. Tango Therapeutics Announces First Patient Dosed in TNG260 Phase 1/2 Trial in Patients With STK11-Mutant Cancers

TNG260 is a first-in-class inhibitor of the CoREST complex . “The TNG260 phase 1/2 clinical trial is the first trial to use genetic patient selection in combination with checkpoint inhibitor therapy to reverse the cancer-specific immune evasion caused by STK11 loss of function mutations. Based on preclinical xenograft studies and retrospective clinical analyses, the majority of STK11-mutant cancers are thought to have primary resistance to checkpoint inhibition. The CoREST complex plays a central role in regulating immunomodulatory signaling in STK11-mutant cancers. In preclinical studies, TNG260 reverses the immune evasion effect of STK11 loss-of-function mutations, restoring sensitivity to an anti-PD-1 antibody, inducing complete remissions in the majority of animals and creating immune memory that prevents re-implantation and regrowth of the tumor. In April 2023, the U.S. Food and Drug Administration granted Fast Track designation for TNG260 in combination with an anti-PD-1 antibody for the treatment of patients with previously treated advanced non-small cell lung cancer with STK11-mutations.
